The urinary tract
A urinary tract infection is the most common condition affecting the urinary tract, which comprises the ureter, the bladder, the urethra and the renal pelvis. Men, women and children can all develop a urinary tract infection. Women in particular are frequently affected by this type of infection, as the short urethra facilitates the entry of pathogens. However, older men are also affected if an enlarged prostate prevents the complete emptying of the bladder. In healthy people, urine is sterile (contains no microorganisms). Regular and complete emptying of the bladder is one of the most important measures for ensuring the sterility of urine. The infection usually begins in the urethra. It can then spread to the upper urinary tract and even reach the kidneys. The symptoms of an infection can vary: a burning sensation, a strong and frequent urge to urinate and, in more severe cases, pain in the lumbar region, fever and chills. In addition, the urine may be cloudy or have a strong odour. The URINARY TRACT INFECTIONS TEST consists of rigid plastic strips on which several separate reagent fields are applied. The test is intended for the qualitative and semi-quantitative detection of the following analytes in urine: leukocytes, blood, nitrites and protein.
Test principle
- Leucocytes: This test detects the presence of granulocyte esterases. The esterases hydrolyse a derivatised pyrazole amino acid ester, thereby releasing hydroxylpyrazole. This pyrazole then reacts with a diazonium salt to produce a beige-pink to violet colour.
- Blood: This test is based on a peroxidase-like activity of haemoglobin, which catalyses the reaction between diisopropylbenzene dihydroperoxide and 3,3’,5,5’-tetramethylbenzidine. The resulting colour ranges from orange through green to dark blue.
- Nitrite: This test is based on the conversion of nitrate to nitrite by Gram-negative bacteria in the urine. In an acidic environment, nitrite in the urine reacts with p-arsanilic acid to form a diazonium compound. The diazonium compound, in turn, binds to 1N-(1-naphthyl)-ethylenediamine to produce a pink colour.
- Protein: This reaction is based on the phenomenon known as the ‘protein error’ of pH indicators, whereby a strongly buffered indicator changes colour in the presence of proteins (anions), as the indicator donates hydrogen ions to the protein. The colours range from yellow to yellow-green for negative results and from green to green-blue for positive results.

Instructions
- Collect your first morning urine in a dry, clean container;
- Remove the test strip from the foil pouch;
- Dip the test strip into the sample for 1–2 seconds;
- Wait 2 minutes to read the result.
Interpretation of results:
- Negative: No leukocytes, blood, nitrites or proteins were detected in the urine.
- Positive for leukocytes: Leukocytes have been detected in the urine. It is recommended that you consult your doctor.
- Positive for blood: Blood has been detected in the urine. It is recommended that you consult your doctor.
- Positive for nitrites: Nitrites have been detected in the urine. It is recommended that you see your doctor.
- Positive for protein: Protein has been detected in the urine. It is recommended that you see your doctor.
